Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000218.3(KCNQ1):c.629T>C (p.Met210Thr), citing Ambry Variant Classification Scheme 2023: The p.M210T variant (also known as c.629T>C), located in coding exon 4 of the KCNQ1 gene, results from a T to C substitution at nucleotide position 629. The methionine at codon 210 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6500 samples (13000 alleles) with coverage at this position. This amino acid position is not conserved on species alignment. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
