This variant lies in the SMAD3 gene (transcript NM_005902.4) at coding-DNA position 850, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 284 with glutamine — a missense variant. Submitter rationale: The p.E284Q variant (also known as c.850G>C), located in coding exon 6 of the SMAD3 gene, results from a G to C substitution at nucleotide position 850. The glutamic acid at codon 284 is replaced by glutamine, an amino acid with some highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6500 samples (13000 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
