NM_000257.4(MYH7):c.3320A>C (p.Lys1107Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1107T variant (also known as c.3320A>C), located in coding exon 24 of the MYH7 gene, results from an A to C substitution at nucleotide position 3320. The lysine at codon 1107 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
