This variant lies in the CBS gene (transcript NM_000071.3) at coding-DNA position 394, where C is replaced by A; at the protein level this means replaces arginine at residue 132 with serine — a missense variant. Submitter rationale: The p.R132S variant (also known as c.394C>A), located in coding exon 3 of the CBS gene, results from a C to A substitution at nucleotide position 394. The arginine at codon 132 is replaced by serine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be possibly damaging but tolerated by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
