This variant lies in the FBN2 gene (transcript NM_001999.4) at coding-DNA position 7250, where G is replaced by T; at the protein level this means replaces glycine at residue 2417 with valine — a missense variant. Submitter rationale: The p.G2417V variant (also known as c.7250G>T), located in coding exon 57 of the FBN2 gene, results from a G to T substitution at nucleotide position 7250. The glycine at codon 2417 is replaced by valine, an amino acid with dissimilar properties. This variant was not reported in population-based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
