This variant lies in the GFAP gene (transcript NM_002055.5) at coding-DNA position 403, where G is replaced by A; at the protein level this means replaces alanine at residue 135 with threonine — a missense variant. Submitter rationale: Variant summary: GFAP c.403G>A (p.Ala135Thr) results in a non-conservative amino acid change located in the Intermediate filament, rod domain (IPR039008) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 249976 control chromosomes (gnomAD).
