Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004982.4(KCNJ8):c.6G>T (p.Leu2Phe), citing Ambry Variant Classification Scheme 2023: The p.L2F variant (also known as c.6G>T), located in coding exon 1 of the KCNJ8 gene, results from a G to T substitution at nucleotide position 6. The leucine at codon 2 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
