Submitter rationale: Variant summary: GLI2 c.1445A>C (p.Lys482Thr) results in a non-conservative amino acid change located in the Zinc finger C2H2-type domain (IPR013087) of the encoded protein sequence. Three of three in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 251396 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1445A>C has been reported in the literature in one individual affected with clinically suspected Maturity-Onset Diabetes of the Young (Tosur_2021).
