NM_145239.3(PRRT2):c.741del (p.Ser248fs) was classified as Pathogenic for Episodic kinesigenic dyskinesia 1 by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PRRT2 c.741delC (p.Ser248AlafsX65) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 244258 control chromosomes. c.741delC has been reported in the literature in individuals affected with Episodic Kinesigenic Dyskinesia 1 and Infantile convulsions (e.g. Cloarec_2012). These data indicate that the variant is likely associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 23077017). No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as pathogenic.
