Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000019.9:g.(17679523_17683291)_(17693967_?)del, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exons 6-12 in the COLGALT1 gene. A presumed nomenclature of c.(829+1_830-1)_(*1714_?)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a large deletion including the last exon in the COLGALT1 gene where loss of function has not been established as a mechanism of disease. The variant was absent in 21694 control chromosomes (gnomAD, Structural Variants dataset). To our knowledge, no occurrence of c.(829+1_830-1)_(*1714_?)del in individuals affected with Brain Small Vessel Disease 3 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
