Uncertain significance for CARD11-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_032415.7(CARD11):c.1754+1G>T, citing ACMG Guidelines, 2015: The CARD11 c.1754+1G>T variant is predicted to disrupt the GT donor site and interfere with normal splicing. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Loss of function is not an established mechanism of CARD11-related disease. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
