Submitter rationale: The p.H1269R variant (also known as c.3806A>G), located in coding exon 48 of the COL3A1 gene, results from an A to G substitution at nucleotide position 3806. The histidine at codon 1269 is replaced by arginine, an amino acid with highly similar properties. Based on data from ExAC, the G allele has an overall frequency of less than 0.01% (1/105450). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
