Uncertain significance for ALMS1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_001378454.1(ALMS1):c.10312A>G (p.Ile3438Val), citing ACMG Guidelines, 2015. This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 10312, where A is replaced by G; at the protein level this means replaces isoleucine at residue 3438 with valine — a missense variant. Submitter rationale: The ALMS1 c.10315A>G variant is predicted to result in the amino acid substitution p.Arg3439Gly. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.058% of alleles in individuals of Latino descent in gnomAD (http://gnomad.broadinstitute.org/variant/2-73786203-A-G). Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
