Uncertain significance for CHEK1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_001114122.3(CHEK1):c.467G>A (p.Arg156Gln), citing ACMG Guidelines, 2015: The CHEK1 c.515G>A variant is predicted to result in the amino acid substitution p.Arg172Gln. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.14% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-125503100-G-A). Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
