NM_005902.4(SMAD3):c.376C>T (p.His126Tyr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H126Y variant (also known as c.376C>T), located in coding exon 2 of the SMAD3 gene, results from a C to T substitution at nucleotide position 376. The histidine at codon 126 is replaced by tyrosine, an amino acid with similar properties. This variant has been reported in a thoracic aortic aneurysm and dissection (TAAD) cohort; however, clinical details were limited (Arnaud P et al. Genet. Med., 2019 Feb; Hostetler EM et al. J. Med. Genet., 2019 Apr;56:252-260). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
