This variant lies in the UNC13A gene (transcript NM_001080421.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 53, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The UNC13A c.53-2A>G variant is predicted to disrupt the AG splice acceptor site and interfere with normal splicing. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Loss of function is not an established mechanism of UNC13A-related disease. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
