To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Several nearby amino acid changes (p.Val216Asp, p.Arg219Ser, and p.Arg219Cys) have been reported in individuals with brancio-oculo-facial syndrome (Meshcheryakova et al. 2015. PubMed ID: 25590586; Milunsky. 2011. PubMed ID: 21204207). Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
