Submitter rationale: The NOTCH1 c.1556-8G>A variant is predicted to interfere with splicing. This variant is predicted to weaken the canonical splice acceptor site and create a cryptic splice acceptor site based on available splicing prediction programs (Alamut Visual Plus v1.6.1). However, such computer prediction programs are imperfect. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare.
