Uncertain significance for BACH2-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_021813.4(BACH2):c.931C>T (p.Arg311Trp), citing ACMG Guidelines, 2015. This variant lies in the BACH2 gene (transcript NM_021813.4) at coding-DNA position 931, where C is replaced by T; at the protein level this means replaces arginine at residue 311 with tryptophan — a missense variant. Submitter rationale: The BACH2 c.931C>T variant is predicted to result in the amino acid substitution p.Arg311Trp. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0080% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/6-90660894-G-A), which may be too common to be causative. Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
