Uncertain significance for Patterned macular dystrophy 3; Pigmentary retinopathy; Blurred vision — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_001243925.2(MAPKAPK3):c.900C>G (p.Asn300Lys), citing ACMG Guidelines, 2015. This variant lies in the MAPKAPK3 gene (transcript NM_001243925.2) at coding-DNA position 900, where C is replaced by G; at the protein level this means replaces asparagine at residue 300 with lysine — a missense variant. Submitter rationale: The missense variant p.N300K in MAPKAPK3 (NM_001243925.2) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.N300K variant is novel (not in any individuals) in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. There is a moderate physicochemical difference between asparagine and lysine. The p.N300K missense variant is predicted to be damaging by both SIFT and PolyPhen2. The nucleotide c.900 in MAPKAPK3 is predicted conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance
