This sequence change creates a premature translational stop signal (p.Tyr266*) in the CYP11B1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CYP11B1 are known to be pathogenic (PMID: 8506298, 26476331). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CYP11B1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
