NM_000548.5(TSC2):c.3666C>G (p.Asp1222Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1222E variant (also known as c.3666C>G), located in coding exon 30 of the TSC2 gene, results from a C to G substitution at nucleotide position 3666. The aspartic acid at codon 1222 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
