Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.8571_8597dup (p.Val2865_Lys2866insAsnArgSerSerSerArgLeuGlyVal), citing Ambry Variant Classification Scheme 2023: The c.8574_8600dup27 variant (also known as p.N2858_V2866dup), located in coding exon 10 of the ALMS1 gene, results from an in-frame duplication of 27 nucleotides at nucleotide positions 8574 to 8600. This results in the duplication of 9 extra residues (NRSSSRLGV) between codons 2858 and 2866. This amino acid region is not well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
