NM_144573.4(NEXN):c.1004G>A (p.Arg335Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R335K variant (also known as c.1004G>A), located in coding exon 8 of the NEXN gene, results from a G to A substitution at nucleotide position 1004. The arginine at codon 335 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
