Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000890.5(KCNJ5):c.365G>A (p.Gly122Asp), citing Ambry Variant Classification Scheme 2023: The p.G122D variant (also known as c.365G>A), located in coding exon 1 of the KCNJ5 gene, results from a G to A substitution at nucleotide position 365. The glycine at codon 122 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
