NM_017946.4(FKBP14):c.133G>T (p.Gly45Trp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G45W variant (also known as c.133G>T), located in coding exon 1 of the FKBP14 gene, results from a G to T substitution at nucleotide position 133. The glycine at codon 45 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
