Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4370T>C (p.Met1457Thr), citing Ambry Variant Classification Scheme 2023: The p.M1411T variant (also known as c.4232T>C), located in coding exon 39 of the KIF1B gene, results from a T to C substitution at nucleotide position 4232. The methionine at codon 1411 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
