NM_001365951.3(KIF1B):c.3499T>C (p.Tyr1167His) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1121H variant (also known as c.3361T>C), located in coding exon 29 of the KIF1B gene, results from a T to C substitution at nucleotide position 3361. The tyrosine at codon 1121 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
