Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4599C>G (p.Ser1533Arg), citing Ambry Variant Classification Scheme 2023: The p.S1487R variant (also known as c.4461C>G), located in coding exon 40 of the KIF1B gene, results from a C to G substitution at nucleotide position 4461. The serine at codon 1487 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
