Uncertain significance — the classification assigned by Ambry Genetics to NM_018975.4(TERF2IP):c.502G>C (p.Glu168Gln), citing Ambry Variant Classification Scheme 2023: The p.E168Q variant (also known as c.502G>C), located in coding exon 1 of the TERF2IP gene, results from a G to C substitution at nucleotide position 502. The glutamic acid at codon 168 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
