Uncertain significance — the classification assigned by Ambry Genetics to NM_018975.4(TERF2IP):c.362G>A (p.Gly121Asp), citing Ambry Variant Classification Scheme 2023: The p.G121D variant (also known as c.362G>A), located in coding exon 1 of the TERF2IP gene, results from a G to A substitution at nucleotide position 362. The glycine at codon 121 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
