NM_001166108.2(PALLD):c.3171G>C (p.Gln1057His) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1040H variant (also known as c.3120G>C), located in coding exon 17 of the PALLD gene, results from a G to C substitution at nucleotide position 3120. The glutamine at codon 1040 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
