Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_002230.4(JUP):c.800C>A (p.Ala267Asp), citing Ambry Variant Classification Scheme 2023: The p.A267D variant (also known as c.800C>A), located in coding exon 4 of the JUP gene, results from a C to A substitution at nucleotide position 800. The alanine at codon 267 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
