NM_002691.4(POLD1):c.401G>C (p.Gly134Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G134A variant (also known as c.401G>C), located in coding exon 3 of the POLD1 gene, results from a G to C substitution at nucleotide position 401. The glycine at codon 134 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
