Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003924.4(PHOX2B):c.269G>A (p.Gly90Asp), citing Ambry Variant Classification Scheme 2023: The p.G90D variant (also known as c.269G>A), located in coding exon 2 of the PHOX2B gene, results from a G to A substitution at nucleotide position 269. The glycine at codon 90 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
