This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 16 through coding-DNA position 18, replacing the reference sequence with ACA; at the protein level this means replaces alanine at residue 6 with threonine — a missense variant. Submitter rationale: The c.16_18delGCCinsACA variant (also known as p.A6T), located in coding exon 1 of the SDHB gene, results from an in-frame deletion of GCC and insertion of ACA at nucleotide positions 16 to 18. This results in the substitution of the alanine residue for a threonine residue at codon 6, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
