Uncertain significance — the classification assigned by Ambry Genetics to NM_006506.5(RASA2):c.1810G>T (p.Val604Leu), citing Ambry Variant Classification Scheme 2023: The p.V604L variant (also known as c.1810G>T), located in coding exon 18 of the RASA2 gene, results from a G to T substitution at nucleotide position 1810. The valine at codon 604 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
