NM_000492.4(CFTR):c.2145A>G (p.Gln715=) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2145A>G variant (also known as p.Q715Q), located in coding exon 14 of the CFTR gene, results from an A to G substitution at nucleotide position 2145. This nucleotide substitution does not change the glutamine at codon 715. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
