NM_001184.4(ATR):c.7217T>A (p.Leu2406His) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L2406H variant (also known as c.7217T>A), located in coding exon 43 of the ATR gene, results from a T to A substitution at nucleotide position 7217. The leucine at codon 2406 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
