NM_198578.4(LRRK2):c.100A>T (p.Thr34Ser) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T34S variant (also known as c.100A>T), located in coding exon 1 of the LRRK2 gene, results from an A to T substitution at nucleotide position 100. The threonine at codon 34 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
