Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.1932A>C (p.Lys644Asn), citing Ambry Variant Classification Scheme 2023: The p.K644N variant (also known as c.1932A>C), located in coding exon 10 of the BRCA2 gene, results from an A to C substitution at nucleotide position 1932. The lysine at codon 644 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
