Uncertain significance — the classification assigned by Ambry Genetics to NM_000903.3(NQO1):c.418A>T (p.Ser140Cys), citing Ambry Variant Classification Scheme 2023: The p.S140C variant (also known as c.418A>T) is located in coding exon 5 of the NQO1 gene. The serine at codon 140 is replaced by cysteine, an amino acid with dissimilar properties. This change occurs in the first base pair of coding exon 5. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
