Uncertain significance — the classification assigned by Ambry Genetics to NM_052947.4(ALPK2):c.1000G>C (p.Asp334His), citing Ambry Variant Classification Scheme 2023: The p.D334H variant (also known as c.1000G>C), located in coding exon 3 of the ALPK2 gene, results from a G to C substitution at nucleotide position 1000. The aspartic acid at codon 334 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
