Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.1681G>C (p.Ala561Pro), citing Ambry Variant Classification Scheme 2023: The p.A515P variant (also known as c.1543G>C), located in coding exon 16 of the KIF1B gene, results from a G to C substitution at nucleotide position 1543. The alanine at codon 515 is replaced by proline, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
