NM_001365951.3(KIF1B):c.3970G>C (p.Glu1324Gln) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1278Q variant (also known as c.3832G>C), located in coding exon 35 of the KIF1B gene, results from a G to C substitution at nucleotide position 3832. The glutamic acid at codon 1278 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
