NM_001148.6(ANK2):c.2743T>A (p.Tyr915Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2743T>A (p.Y915N) alteration is located in exon 25 (coding exon 25) of the ANK2 gene. This alteration results from a T to A substitution at nucleotide position 2743, causing the tyrosine (Y) at amino acid position 915 to be replaced by an asparagine (N). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
