Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.6344T>C (p.Leu2115Ser), citing Ambry Variant Classification Scheme 2023: The p.L2115S variant (also known as c.6344T>C), located in coding exon 15 of the APC gene, results from a T to C substitution at nucleotide position 6344. The leucine at codon 2115 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
