NM_017802.4(DNAAF5):c.2434G>A (p.Val812Ile) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V812I variant (also known as c.2434G>A), located in coding exon 13 of the DNAAF5 gene, results from a G to A substitution at nucleotide position 2434. The valine at codon 812 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
