Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.2661A>T (p.Lys887Asn), citing Ambry Variant Classification Scheme 2023: The p.K887N variant (also known as c.2661A>T), located in coding exon 22 of the BUB1 gene, results from an A to T substitution at nucleotide position 2661. The lysine at codon 887 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
