Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.2225G>A (p.Cys742Tyr), citing Ambry Variant Classification Scheme 2023: The p.C742Y variant (also known as c.2225G>A), located in coding exon 10 of the ATR gene, results from a G to A substitution at nucleotide position 2225. The cysteine at codon 742 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
